Peptide-5 is a synthetic polypeptide with the amino acid sequence glycine-proline-arginine-proline-alanine. It acts as a competitive antagonist of the AChR by binding to the AChR and preventing acetylcholine (ACh) from binding to it, thereby causing receptor blockade.
Sodium ions cannot be released on the postsynaptic membrane, which reduces the generation of electrical signals and, consequently, decreases muscle contraction. Studies have shown that in vitro application of Pentapeptide-3 is highly effective: it immediately reduces muscle activity by 71%, with the effect gradually declining to 58% after two hours. After four consecutive weeks of use, the depth of wrinkles was reduced by 49%.2 This suggests that Pentapeptide-3 can effectively inhibit the release of the neurotransmitter acetylcholine, thereby blocking—or even interrupting—the very first step of signal transmission.
